SYNTHESIS OF N – [(2, 7 – DICHLORO – 10H – PHENOTHIAZINE – 10 YL) METHYL] – N – (4 – METHYLPHENYL) AMINE AS A NOVEL NOOTROPIC DRUG

ABSTRACT
If the mental health is not proper then the physical health has no value. Hence to obtain better Nootropic (Learning and Memory enhancing) Activity we have synthesized two series of some fused bicyclo phenothiazine compounds. In first series, compounds (N1-N5, N10-N11) have been synthesized by treating formaldehyde, aryl amine with 7, 8, 9 substituted -1- carboxyl phenothiazine in presence of hydrochloric acid by Mannich reaction. 2-carboxy-2’, 3’ and 4’ substituted diphenylamine have been synthesized by the reaction between 2- chlorobenzoic acid with 2, 3, 4 substituted aniline respectively in the presence of anhydrous K2CO3 and CuO by Ullmann reaction. In second series the starting material 2 - chlorobenzoic acid was replaced by m-dichlorobenzene and the titled compounds (N6-N9 and N12) were synthesized. Titled compounds were recrystallized and obtain in good yield. The melting points were taken in open capillary and are uncorrected. Synthesized compounds were characterized by spectral analysis. Compounds N7 were evaluated for Nootropic Activity by using Cook’s Pole Climbing apparatus. Activity of N7 were compared with standard Piracetam and has shown significant activity than the standard hence it can be utilized in treating diseases with memory deficits like senile Dementia and Alzheimer’s disease.
