HAEMATOLOGICAL AND HISTOPATHOLOGICAL ALTERATIONS DUE TO ISOLATED AND CROWDING STRESS AND THE ACTION OF SOME ANTISTRESS AGENTS (MEDICINAL AND HERBAL) ON THE MALE ALBINO RATS

ABSTRACT
Stress, constitutes a major threat to psychology. Mood, anxiety and sleep disorders are prevalent and psychiatric conditions developed by various stress. Such as crowding and isolation stress. The present study was designed to investigate the adverse effect of crowding and isolation stress and the effect of alprazolam group medicine which are widely used in treating anxiety; and protective effects of Withania somnifera against the adverse effect induced by the drugs and stress on haematological and histophysiological alterations.The Withania somnifera were frequently used by the medicine men of the Rajbanshi
community District Coochbehar , W.B, India. Considering its efficacy Withania somnifera was used on the male albino rat during stressful condition. The experimental study was conducted with healthy male Albino rats (weighing 90-120gm) the experiment was carried out according to the guidelines and prior approval of Institutional Animal Ethics Committee. Animals were divided into seven groups, e.g., control and other six experimental groups (group A, B, C, D, E and F) were treated with alprazolam and the extract of a herbal medicine, the root extract of Withania Somnifera during the isolated and crowding stress. The experiment was carried out for 14 days. The haematological assays showed a significant reduction in packed cell volume, haemoglobin, total erythrocytes count and Alprazolam was found to induce a significant increase in neutrophil count and a significant decrease in lymphocytes in group C and group D in comparison to the other groups., Histophysiological changes were noticed significantly. In group C and group D, adrenal showed severe zona fasciculata congestion and mild congestion of central vein which were infiltrated with mononuclear cells. Disorganization of cords and moderate to severe necrosis were noticed. In kidney, glomeruli revealed degeneration of tubules and haemorrhages in interstitial tubular spaces. The groups E and F animals showed significant improvement in all parameters in comparison to the groups C and D which may be due to effects of Withania somnifera.
