AN ULTRA-PERFORMANCE LIQUID CHROMATOGRAPHY-QUADRUPOLE TIME-OF-FLIGHT MASS SPECTROMETRIC METHOD FOR IN VITRO METABOLISM STUDY OF PHENACETIN IN MOUSE LIVER

ABSTRACT
An ultra performance liquid chromatography coupled to quadrupole time-of-flight analyzers (UPLC-QTOF-MS) for screening, identification, ad quantification of phenacetin and paracetamol was developed. The parameters of UPLC-QTOF-MS were investigated and optimized. Linearity, sensitivity, repeatability, and accuracy of the method were validated. An accurate-mass database containing retention time, calculated mass, mass error, elemental composition, and double bond equivalent for the formula was established. Screening, identification, ad quantification of phenacetin and paracetamol in mouse liver homogenates were carried out. The validated method was successfully used for in vitro metabolism study.
