Abstract

INFLUENCE OF MITOCHONDRIAL DNA ON SODIUM CHLORIDE INDUCED APOPTOTIC DNA DAMAGE AND OXIDATIVE STRESS IN MICE

ABSTRACT

This study was aimed to investigate the influence of mitDNA damage on NaCl induced apoptosis and oxidative stress in mice. One fixed dose (10 mL/kg body weight) of saline different concentrations (0.9, 3, or 5%) was injected into the abdominal cavity of mice every two days for a total of 6 injections and euthanized 24 h after the last injection. The observed significant elevations in apoptotic fractions using neutral comet assay and appearance of smeared pattern of genomic DNA running on agarose gel evidenced the apoptotic DNA damage inductions by NaCl saline in a concentration dependent manner. This could be attributed to the observed concurrent high mutations incidence in D-loop of mitDNA that therapy increased the free radical generations by saline as indicated by the observed significant elevations in malondialdehyde level and decreases in the antioxidant reduced glutathione level and catalase activity in NaCl treated groups. Conclusion: mutations in mitDNA D loop fragments takes part in the observed NaCl induced apoptotic DNA damage and oxidative stress.
