Abstract

“DYNAMICS OF ANTIBODY GENERATION IN WHITE LEGHORN CHICKENS AGAINST ROTATEQ® VACCINE – A PASSIVE THERAPY APPROACH”

ABSTRACT

Gastroenteritis is one of the major concerns globally. In humans, this infection becomes drastic in nature due to Rotavirus. This rotavirus infection becomes severe in children below 5 years of age. An impeccable and the best treatment for this infection, caused by rotavirus is the passive therapy approach with IgY to preclude its severity. The present study is to elevate antibodies against Rotavirus in chickens immunized with RotaTeq® vaccine and characterization of anti-rotavirus IgY. The purified and freeze dried anti-rotavirus IgY antibodies are used for certain dynamics studies and its titration aspects. The titre of IgY in the final preparation was determined to be 1:100000 on the 49th titre day upon performing titration techniques with ELISA. The protein concentration of the purified antibodies was also estimated and it was found to remain increasing between 16.406 mg/ml of IgY extract during the entire study period. The purity of IgY extracts was determined by SDS-PAGE and Column Chromatography techniques where the bands are observed in the test lane and standard IgY were similar. It indicates effective purity and also confirms the molecular mass of IgY as 180KD, extracted from the egg yolk. The exploration of this study also deals with a Passive Therapy approach in children with rotavirus infections who are not vaccinated by birth. Infants under this category who are neither vaccinated nor hold immunity naturally can be treated to cure the infections caused by rotavirus. The therapeutic efficacy and a passive approach of IgY using a small animal model could be achieved in further research uptake.
