Abstract

EFFECT OF PHYSICAL AND NUTRITIONAL PARAMETERS FOR OPTIMUM BIOSURFACTANT PRODUCTION BY STREPTOMYCES MATENSIS (NBRC 12889T)

ABSTRACT

Aim: To evaluate the effect of physical and nutritional parameters by response surface methodology using Box-Benkhen design to optimize production of biosurfactant by the actinomycetes Streptomyces matensis (NBRC 12889T). Methods and Results: Biosurfactant was produced by actinomycetes strain NBRC 12889T, isolated from soil contaminated with poultry litter, in seven day submerged fermentation medium. Using olive-oil, NaNO3, level-of-inoculum concentration as independent variables, biosurfactant production was depicted adopting a statistically second-order quadratic regression model. Optimum values for these independent variables to achieve the maximum predicted value of 425.656 μg/ml were derived as olive oil (3.021% v/v), NaNO3 (0.102% w/v) and level-of-inoculum (10.761% v/v). Conclusions: Closeness of predicted values by statistical analysis and experimental results suggested reliability of regression model adopted. A 9.366% increase in biosurfactant production during experiments, 465.524 μg/ml compared to predicted 425.656 μg/ml, further confirmed significant positive effect of optimized physical and nutritional parameters of culture medium on biosurfactant production. Significance and impact of the Study: This study proved the effectiveness of using actinomycetes as bio-producer and olive oil as sole carbon source in fermentation medium, and verified the benefit of using optimized physical and nutritional parameters for improved biosurfactant production. Further, isolated actinomycetes strain Streptomyces matensis (NBRC 12889T) was found useful for bioremediation and anticancer activity
