Abstract

ANTIOXIDANT ACTIVITY OF DEHYDROZINGERONE ON STZ AND L-NAME INDUCED OXIDATIVE STRESS IN EXPERIMENTAL RAT MODEL

ABSTRACT

This study evaluates the antioxidant effects of dehydrozingerone (DZ) against oxidative stress in Streptozotocin (STZ) and N-nitro-L-arginine methyl ester (L-NAME) induced diabetic hypertension in male Wistar rats. Hypertension was prompted in adult Wistar rats, weighing 180–220g, by oral administration of the L-NAME (40 mg/kg bodyweight/day) dissolved in drinking water for 4 weeks and diabetes was invoked by a single intraperitoneal injection of 40 mg/kg of streptozotocin (STZ). Rats were given intraperitoneal injection of DZ (100 mg/kg bodyweight). The diabetic hypertensive rats revealed a significant increase (P<0.05) in the levels of thiobarbituric acid reactive substances (TBARS) and lipid hydroperoxides (LOOH) in plasma and tissues. Enzymic antioxidants such as superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) in erythrocytes and tissues, and non-enzymic antioxidants such as reduced glutathione, vitamin C and E in plasma tissues were also significantly decreased. Administrations of DZ suppress the oxidative stress and improved the levels of enzymic and non enzymic antioxidant. Our results suggested that DZ exerts antioxidant effects against STZ and L-NAME induced oxidative stress in diabetic hypertensive Wistar rats.
