Abstract

ARCHAEOSOMES: A NOVEL LIPID BASED DRUG DELIVERY SYSTEM

ABSTRACT

Archaeosome (liposomes comprised of glycero-lipids of archaea) - a novel lipid based oral drug delivery system was developed, which is more stable and possesses adjuvant activity that make archaeosome more useful in vaccine / gene delivery system. Archaeosome are also used for encapsulation of various biologically active molecules like DNA, peptide, antigen, etc. the present study demonstrates the structural features, adjuvant activity, preparation, characterization, stability, toxicity and applications of archaeosomes. Liposomes are lipid-based nano carrier system1 composed of natural or synthetic ester phospholipids(2) and used for encapsulation, targeting and controlled release technologies .the materials with different solubilities can be incorporated into the liposomes and delivered at the required site, both inside as well as outside the body, in- vitro.(1) Archaeosomes are nothing but a liposomes in 200nm size range, which are prepared from ether lipids extracted from various Archaeobacteria Archaeosomes(liposomes comprised of glycerolipids of Arcaea)(5) are lipid vesicles (6) and their membranes are constituted of ether linked isoprenoid phytanyl core lipid(7) or purified lipid sub fractions9.Archaeosomes constitute a novel family of liposomes(9) made with one or more of the fully saturated bipolar tetra ether lipids, which exerts a higher stability in comparison with conventional lipids to several conditions such as high temperature, alkaline or acidic pH, presence of phospholipases, bile salts and serum media.(10) An agent that stimulates the immune system and increases the response to a vaccine, without having any specific antigenic effect in itself is usually referred to as an adjuvant (latin-adiuvare, meaning to help or aid) in immunology. Potent adjuvant activity of archaeosomes administered through parentral route(11) is useful for combination vaccine and correlate to recruitment and activation of macrophages and dendrite cells In-vivo
