INVESTIGATION OF TAMARINDUS INDICA LINN. HYDROMETHANOLIC LEAF EXTRACT AGAINST GLUCOSE INDUCED CATARACTOGENESIS

ABSTRACT
Cataract is the opacification of lens often associated with old age and is a major complication of diabetes mellitus because higher glycosylated hemoglobin levels are significantly associated with increased risk of cataract. The medicinal plant Tamarindus indica Linn. was proved its antioxidant activity by using in vitro & in vivo studies. This current study is investigating of in vitro anticataract activity of Tamarindus indica Linn. against glucose-induced cataractogenesis by using isolated goat lenses. Isolated goat lenses were incubated in artificial aqueous humor (TC 199 medium) containing 55mM glucose (cataractogenesis) and Tamarindus indica Linn. extract in different concentrations kept at room temperature for 72 h and renewed with fresh solution for every 24h. Lenses remain clear during the initial 24h but form shell like opacity around the nucleus by 48h. Opacification becomes more peripheral and wide spread after 72h. Biochemical parameters were studied in the lens homogenate prepared in Tris buffer, which are end metabolites like malondialdehyde (MDA), lipid peroxidase and proteins, enzymatic and non enzymatic antioxidants. In results the cataractous lenses showed higher MDA (P<0.001), and water-soluble protein content and decreased in antioxidants. Lenses treated with Tamarindus indica Linn. Leaf extract in concentrations of 100, 200µg/ml showed decreased MDA, higher protein (total proteins) content, increased antioxidants and prevented formation and progress of cataract by glucose, as evidenced by biochemical parameters.
