Abstract

PREPARATION AND EVALUATION OF MULTIPLE UNIT PELLET SYSTEM OF METRONIDAZOLE FOR COLON TARGETING

ABSTRACT

Objective: The purpose of study was to development of Metronidazole colon targeted pellets to make maximum amount drug at targeted site which minimize the conventional dose, side effects, toxic effects and avoid hepatic metabolism. Experimental Work: Metronidazole pellets were prepared by Extrusion-spheronization process by using MCC and different super-disitegrants such as SSG, Crospovidone and Kyron-314. The powders and pellets were subjected to evaluation of various before coating and after coating parameters such as Friability, Sphericity, Aspect ratio, Yield, % drug content, Avg. Particle size and in-vitro dissolution study. Result: Results were found to be within the acceptable limits. The results of in vitro release data indicate that desirable drug release was obtained through double layer coated formulation. Based on that result, optimized formulation was M7. Stability study of optimized formulation showed that Metronidazole pellets were stable at accelerated environment condition of 40oC/75 % RH. Conclusion: Friability, Sphericity, Aspect ratio and yield can be achieved in acceptable range by using optimum level of water and spheronization speed. Also the desirable release can be achieved by using Eudragit RS100 (inner coat) and Eudragit S100 (outer coat).
