Abstract

FORMULATION, OPTIMIZATION AND EVALUATION OF CEPHALEXIN FLOATING TABLETS

ABSTRACT

The purpose of this research was to formulation, optimization and evaluation of cephalexin floating tablet by using the hydrophilic polymer hydroxypropyl methyl cellulose and gas generating agent sodium bicarbonate and tartaric acid. Cephalexin is a semi synthetic cephalosporin antibiotic having dose 1g daily, in divided doses and well absorbed in stomach. A 32 factorial design was applied systematically to optimize the drug release profile. The amount of tartaric acid (X1) and amount of HPMC K 4 M (X2) were selected as independent variables. The times required for 50% drug dissolution (t50%) percentage release at 6 hours (Q6), percentage drug release at 12 hours (Q12) were selected as dependent variables. The results of factorial design indicated that high level HPMC K 4 M and tartaric acid favors preparation of cephalexin floating tablet. The granules were prepared by wet granulation method and evaluated for their granules properties. Tablets were compressed by Karnavati tablet compression machine and evaluated with different parameters like diameter, thickness, average weight, hardness, friability, in vitro buoyancy study, swelling characteristics and stability study. According to stability study it was found that there was no significant change in dissolution profile of optimized formulation.
