Abstract

PHARMACOKINETICS AND TISSUE RESIDUES OF CEPHRADINE IN HEALTHY AND EXPERIMENTALLY SALMONELLA ENTRETIDIS INFECTED BROILER CHICKENS

ABSTRACT

The pharmacokinetic parameters of cephradine (20 mg/kg b.wt.) were studied following intravenous (IV) and oral (PO) (single and repeated) administrations in healthy and experimentally Salmonella entretidis infected broiler chickens. Following a single IV injection of cephradine at a dose of 20 mg/kg b.wt. in healthy chickens, cephradine could be detected therapeutically for 12 h. The serum concentration–time curve of cephradine following IV injection showed that, the drug obeyed a two compartments open model and the elimination half-life (t1/2β) was 2.79 ± 0.047 h, volume of distribution (Vdss) was 328.94±12.27 ml/kg and total body clearance (CLtot) of cephradine was 0.095±0.003 L/h/kg). Following a single PO administration of 20 mg cephradine/kg b.wt. in healthy chickens, the peak serum concentration (Cmax) was 25.65 ± 0.934 μg/ml was achieved at a maximum time (Tmax) of 2.61 ± 0.025 h. The oral bioavailability of cephradine in healthy chickens was 85.91±1.96%. Oral administration of 20 mg cephradine/b.wt. twice daily for five consecutive days in healthy and Salmonella entretidis infected chickens revealed a lower significant serum cephradine concentration after the 1st, 3rd,5th, 7th and 9th doses in Salmonella entretidis infected chickens in comparison to healthy chickens. Cephradine showed accumulative behavior in serum of chickens. Cephradine was assayed in kidney, liver, heart, gizzard, spleen, breast muscle, thigh muscle and skin after 24, 48, 72, 96, 120 and 144 h post last dose. On conclusion, cephradine was useful for treatment of Salmonella entretidis infections in chickens.
