Abstract

FORMULATION, DEVELOPMENT AND EVALUATION OF PROPRANOLOL HYDROCHLORIDE BUCCO-ADHESIVE PATCH

ABSTRACT

The main approach of buccal delivery of desired drug is to avoid first pass effect. Propranolol hydrochloride is a nonselective Beta-adrenergic blocking agent. It is widely used in treatment of hypertension, angina pectoris, and many cardiovascular disorders. Although it is well absorbed from gastrointestinal tract, it’s bioavailability is low (15-23%) because of extensive first pass effect. The objective behind making of attractive drug delivery system is to bypass the extensive first pass effect. the formulation of Propranolol hydrochloride buccal patch was carried out using hydrophilic mucoadhesivepolymer HPMC E-15 and matrix forming polymer Eudragit L-100 by solvent casting method. The patches were evaluated and 32 factorial design was applied using Design Expert®Trial version 7 and analysed the responses for it. 3D plot and contour plot plotted using this software. Various solutions obtained by software analysis and one of it selected as optimized formulation which showed the drug relese 97.04 ± 0.10 with desirability factor 1.00. the optimized formulation followed Higuchi model. So the systemic delivery of Propranolol hydrochloride through bucaal mucosa can be considered as successful.
