MEDIA OPTIMIZATION FOR DOCOSAHEXAENOIC ACID PRODUCTION BY SCHIZOCHYTRIUM SPECIES USING STATISTICAL TECHNIQUE

ABSTRACT
Many optimization techniques are available for the optimization of
fermentation media. Each optimization technique has its own
advantages. In this study, Plackett-Burman design and full factorial
design are used to optimize the media for DHA production. The effect
of various media components such as dextrose, yeast extract, tryptone,
NaCl, MgSO4, CaCl2, KH2PO4, KCl were examined. Dextrose, yeast
extract, tryptone and NaCl were selected as significant factors by
Plackett-Burman design which was further screened by full factorial
design. A central composite design and response surface methodology
were utilized to optimize the screened factors and to increase the
productivity of DHA. A maximum DHA production was achieved 4.48
mg/g (free DHA) with the combination of optimized factors: dextrose 74.92 g/l, tryptone
10.94 g/l and NaCl 5.97 g/l. Validation experiments were also performed to verify the
adequacy and accuracy of the model.
